Elution Buffers (TE, EB, Water)

Final Buffer Concentration
 10 mM Tris-Cl
 1 mM EDTA (TE only)
 pH 8.0
For mixing 200mL of Buffer TE or EB
1

Dissolve 242.28mg Tris Base in 100mL Autoclaved dH20.
Note: Some EB recipes use 5mM Tris-Cl, so you can choose to add instead 121.14g. I have not noticed a difference.

2

Add 58.448mg EDTA (Only if you're making TE - do not add if you wish to make EB)

3

Balance pH to 8.0

4

Filter sterilise or autoclave. Add 15mL to a falcon tube for immediate use and refrigerate the rest.

Water
5

Just autoclave water (MilliQ or RO) in a bunch of small bottles (keep those lids loose!). You can open a new bottle each day to ensure you're always working with the freshest water for all your experiments.
